I've been thinking about this on and off over the years. My number one choice...
Has Graeme Garden ever been approached? He's extremely sharp and quick-witted, very funny, as well as very intelligent, being both a Cambridge graduate and a qualified (though non-practising) doctor. His success on I'm Sorry I Haven't a Clue also shows a very good ability with wordplay. (Incidentally, he also seems to be the only regular Clue performer not to have been on the show.)
Other people I've always wanted to appear over the years, who I think would be very good either because they are particularly witty, charismatic or full of anecdotes (excluding people who have already appeared):
Andy Hamilton
Tom Baker
Sandi Toksvig
Rob Brydon
David Tennant
I always love it when new people appear, and ideally would love to see more new guests appearing and fewer return appearances, at least within the same series.

I wouldn't mind seeing Griff Rhys Jones or Michael Palin on Countdown.
Joseph's suggestion of Joe Pasquale reminded me of Roy 'RIGGHT' Walker, for some reason, and I think he'd be pretty decent in DC. Chalk up another one for Stephen Fry to return too and the slightly less likely selections from me are for Rowan Atkinson, Martin Clunes and Caroline Quentin
